In this work, the PCR method was used to examine polymorphisms in the TAS2R38 gene. That gene encodes a strong receptor for the harmless compound phenylthiocarbamide (PTC), and it is located in the taste bud cells on the tongue. There are people to whom compounds like PTC and 6-N-propylthiouracil (PROP) cause an intense bitter taste, while some people may not feel that bitter taste at all. The reason for this phenomenon is the existence of single nucleotide polymorphisms (SNPs) in that gene. One allele of TAS2R38 encoding that protein is called the PTC taster, while the other allele of this gene does not react with PTC and is called the PTC non-taster. In this work, subjects were first tested for PTC taste sensitivity, then their DNA samples were taken and fragments of their TAS2R38 genes were amplified using the polymerase chain reaction, PCR. The obtained reaction products were treated with restriction endonuclease and the results were analyzed on a gel. Both alleles created fragments of the same length, but the fragment of the allele encoding for the taster has a restriction site for the restriction enzyme SatI (Fnu4HI). Given that each individual has two alleles for that gene, we were able to conclude which of the subjects were homozygous for the PTC receptor (PTC taster), homozygous without the PTC receptor (PTC non-taster), or heterozygous for this trait. The obtained results show a correlation between the sensitivity of individuals to the bitterness of PTC and their genotype of TAS2R38.
