Abstract
Laktoza je disaharid koji se u probavnom sustavu sisavaca razgrađuje posredstvom enzima laktaze. Kao produkti ove reakcije nastaju monosaharidi, glukoza i galaktoza, koje se mogu apsorbirati na sluznici tankog crijeva. Enzimska aktivnost laktaze mijenja se tijekom života pojedinca. Tako se najviša aktivnost laktaze bilježi kod novorođenčadi, a u odraslih se aktivnost laktaze smanjuje što može dovesti do intolerancije na laktozu. S obzirom na sve veći problem intolerancije na laktozu, cilj ovog završnog rada je istražiti molekularno-biokemijsku podlogu te suvremene pristupe u liječenju ove bolesti. Istraživanja su pokazala da je intolerancija na laktozu povezana s polimorfizmom gena lct. Gen Lct koji kodira enzim laktazu nalazi se na drugom kromosomu. Dijagnosticiranje intolerancije se provodi različitim testovima pomoću kojih se utvrđuje radi li se o intoleranciji na laktozu ili nekom drugom gastrointestinalnom problemu. Nakon utvrđivanja dijagnoze, liječenje se zasniva na izbjegavanju unosa namirnica koje sadrže laktozu s ciljem ublažavanja probavnih smetnji pacijenta.
Abstract (english)
Lactose is a disaccharide that in the mammalian digestive system is hydrolyzed by the action of enzyme lactase. This digestion yields monosaccharides, glucose and galactose that are absorbed by the mucus membrane of the small intestine. The lactase activity changes during the lifetime with the highest activity observed in infants. This activity gradually drops during the adulthood which can lead to condition known as lactose intolerance. Considering the growing problem of lactose intolerance, the aim of this thesis is to explore molecular and biochemical basis and modern approaches in the treatment of this disease. Studies have shown that lactose intolerance is associated with the lct gene polymorphism. The lct gene encoding the enzyme lactase is located on the second chromosome. Diagnosis of the lactose intolerance is conducted by various tests that help distinguish lactose intolerance from some other gastrointestinal problems. Once the diagnosis is made, treatment is based on avoiding the lactosecontaining foods in order to alleviate patient’s symptoms.
