Abstract
Razvojem elektronske mikroskopije znanost je napravila golem korak prema brojnim novim, do sada nezamislivim, otkrićima. Transmisijski elektronski mikroskop (TEM) uporabom elektrona umjesto svjetlosnih zraka nudi veću razlučivost i povećanje, no slika koja njome nastaje može biti samo crno-bijela. Najbitniji korak u razvoju TEM-a bio je napredak rezolucije putem pojačanja napona i razvojem naprednijih leća. Sama priprema uzroka za ovu vrstu mikroskopije može biti vrlo zahtjevan i iscrpan proces koji zahtijeva izrazitu vještinu i preciznost. Dok se kod optičkog mikroskopa može proizvesti samo jedna vrsta slike, TEM specifičnim pozicioniranjem leća, nagiba snopa elektrona i promjera otvora leće ima sposobnost stvaranja nekoliko vrsta slike od kojih svaka može prikazati različite strukture.
Abstract (english)
Through the development of electron microscopy, the field of science made a huge step towards new, until then unimaginable, discoveries. Transmission electron microscope (TEM) provides a better display of magnification and resolution by using electrons instead of light rays, but it can only produce a black and white image. The most important step during TEM's development was obtaining higher resolution by increasing the voltage and advancing the lenses. Preparing a sample for this type of microscope alone can be a challenging and draining process that requires extreme skill and precision. While the optical microscope can produce only one type of image, TEM is capable of displaying many different structures due to its ability to adjust specific lens positioning, electron beam tilt and lens aperture diameter.
