Abstract
Preimplantacijska genetička dijagnostika (PGD) je klinički dijagnostički postupak koji je razvijen za determinaciju genetički nasljednih bolesti u preimplantiranim embrijima. Uz primjenu PGD - a u potpomognutoj oplodnji (IVF), smanjio se broj spontanih pobačaja i značajno povećala stopa uspješnih trudnoća kod parova s rizikom od prenošenja bolesti na zametak. Raniji postupci su uključivali invazivnije metode biopsije stanica za testiranje, no uz današnji napredak, rezultati su vidljivi i uz biopsiju polarnog tijela ili trofektodermskih stanica. Prve tehnike PGD - a uključivale su polimeraznu lančanu reakciju (PCR) te fluorescentnu in situ hibridizaciju (FISH) koje su danas zamijenile novi molekularni alati poput DNA mikročipova i sekvenciranja sljedeće generacije (NGS). Nove metode pomogle su prevladati rizik i poboljšati ukupne analize preimplantacijske genetičke dijagnostike. Cilj ovog rada je dati bolji uvid u molekularne tehnike koje se koriste pri PGD - u i bolje razumijevanje polja reproduktivne genetike.
Abstract (english)
Preimplantation genetic diagnosis (PGD) is a clinical diagnostic procedure developed for the determination of genetically inherited diseases in pre-implanted embryos. Introduction of PGD in in vitro fertilization caused the reduction of miscarriages and increasedthe rate of successfully carried pregnancies in couples with higher risk of transmitting a disease to their offspring. Early procedures included more invasive biopsy methods, but with great advancements, the results are visible with polar body biopsy or even trophectoderm cells. First PGD techniques included polymerase chain reaction (PCR) and fluorescent in situ hybridization (FISH), but today they are replaced with new molecular tools such as DNA array and next-generation sequencing. New methods have helped to overcome certain risks and to improve overall PGD analysis. Main aim of this thesis is to give a better insight into molecular techniques that are used in PGD as well as in the field of reproductive genetics.
