Abstract
Rapamicin je makrolid koji ima ulogu u rastu i metabolizmu stanica djelujući na PI3K-PKB/Akt-mTOR (engl. Phophatidylinositol-3-kinase, PI3K; Protein kinase B, PKB; Mammalian target of Rapamycin, mTOR) signalni put koji sudjeluje u brojnim staničnim funkcijama. Glavna meta rapamicina u ovom signalnom putu je mTOR čijom inhibicijom rapamicin djeluje na razvoj bolesti i metabolizam. U ovom radu obrađuje se mehanizam rada rapamicina, njegova uloga u metabolizmu i regulaciji starenja te prednosti i nedostatci korištenja rapamicina pri liječenju bolesti.
Abstract (english)
Rapamycin is a macrolide that plays a role in cell growth and metabolism by acting on the PI3K-PKB/Akt-mTOR (Phophatidylinositol-3-kinase, PI3K; Protein kinase B, PKB; Mammalian target of Rapamycin, mTOR) signalling pathway that participates in numerous cellular functions. The main target of rapamycin in this signalling pathway is mTOR. Inhibition of mTOR by rapamycin affects disease development and metabolism. This thesis explains rapamycin mechanism, its role in metabolism and regulation of aging, and the advantages and disadvantages of using rapamycin in the treatment of diseases.
