Abstract
Razvoj rezistentnih sojeva bakterija predstavlja globalnu opasnost. Stoga istraživanja novih antimikrobnih spojeva često obuhvaćaju ispitivanje potencijala razvoja rezistencije kao i rezistentnih mehanizama. Kvaterne amonijeve soli (engl. quaternary ammonium compounds, QACs) antimikrobni su reagensi širokog spektra djelovanja. Istraživanja su pokazala da je razvoj rezistencije prema QACs najčešće posredovan efluks pumpama Qac. U ovom završnom radu ispitan je antimikrobni potencijal komercijalnih QACs, cetilpiridinijevog klorida i benzalkonijevog bromida, kao i njihov potencijal razvoja rezistencije posredovan Qac efluks pumpama. Ispitivanja su provedena na bakteriji Staphylococcus aureus ATCC 33591 pri čemu su određene minimalne inhibitorne koncentracije (MIK) ovih spojeva bez i u prisustvu inhibitora ATP-sintaze, karbonil cijanid m-klorofenil hidrazona. Dobiveni rezultati upućuju na zaključak da su oba komercijalna QACs spoja supstrati Qac efluks pumpi što potvrđuje objavljene podatke o sve široj incidenciji rezistencije prema ovim spojevima.
Abstract (english)
The development of resistant strains of bacteria represents a global danger. Therefore, research into new antimicrobial compounds often includes testing the potential for resistance development as well as resistance mechanisms. Quaternary ammonium compounds (QACs) are antimicrobial reagents with a wide spectrum of action. Research has shown that the development of resistance to QACs is most often mediated by Qac efflux pumps. In this Final Thesis, the antimicrobial potential of commercial QACs, cetilpiridinium chloride and benzalkonium bromide, as well as their potential for developing resistance mediated by the Qac efflux pumps, was examined. Tests were performed on the bacterium Staphylococcus aureus ATCC 33591, where the minimum inhibitory concentrations (MIC) of these compounds were determined without and in the presence of the ATP-synthase inhibitor, carbonyl cyanide m-chlorophenyl hydrazone. The obtained results point to the conclusion that both commercial QACs compounds are substrates of Qac efflux pumps, which confirms published data on the increasing incidence of resistance to these compounds.
