Abstract
Telomere su krajevi linearnih molekula DNA u eukariotskim kromosomima te su građeni od specifične ponavljajuće sekvence DNA i obloženi zaštitnim proteinima. Njihova je glavna funkcija očuvanje genetičke stabilnosti sveukupnog genoma i zaštita kromosoma od degradacije i fuzija. Zbog prirode replikacijskog mehanizma molekule DNA, pri svakoj replikaciji dolazi do smanjenja duljine telomera, što je povezano s prirodnim starenjem organizma i postupnom degradacijom funkcije kromosoma. Taj prirodni mehanizam otvara široki spektar mogućnosti istraživanja utjecaja skraćenih telomera na zdravlje i životni vijek ljudi. U određenim uvjetima telomere mogu poslužiti kao „molekularni sat“, koji „mjeri“ broj staničnih dioba. Akumulacija iznimno kratkih telomera povećava rizik od razvoja neurodegenerativnih bolesti, raka, kardiovaskularnih bolesti te mnogih drugih poremećaja. Danas je poznata široka skupina ljudskih bolesti povezanih s poremećajima u funkciji i građi telomera, a skraćivanje telomera smatra se i jednim od ključnih čimbenika u starenju organizama.
Abstract (english)
Telomeres are ends of linear DNA molecules in eukaryotic chromosomes which are built from a specific repetitive DNA sequence and coated with protective proteins. Their main function is to preserve the genetic stability of the entire genome and protect chromosomes from degradation and fusion. Due to the nature of the replication mechanism of the DNA molecule, with each replication there is a reduction in the length of the telomeres, which is associated with the gradual deterioration of chromosome function and the natural aging of the organism. This natural mechanism opens up a wide range of possibilities for research into the impact of shortened telomeres on human health and lifespan. Under certain conditions, telomeres can serve as a “molecular clock”, which “measures” the number of cell divisions. The accumulation of extremely short telomeres increases the risk of developing neurodegenerative diseases, cancer, cardiovascular diseases and many other disorders. Today, a wide group of human diseases associated with disorders in the function and structure of telomeres is known, and the shortening of telomeres is considered one of the key factors in the aging of organisms.
