Abstract
Mutacije su trajne promjene u genetičkom materijalu koje nastaju u somatskim ili reproduktivnim stanicama. One mogu biti spontane, kao posljedica pogreške u replikaciji, ili inducirane djelovanjem kemijskih i fizičkih agensa. U kemijske agense ubrajaju se kemijski spojevi koji se metaboliziraju u elektrofilni oblik, alkilirajući agensi, analozi baza, agensi koji premošćuju DNA i metalni ioni. Različitim mehanizmima uzrokuju brojna oštećenja DNA, od kojih su najčešća: supstitucija baza, ugradnja ili uklanjanje baza te stvaranje DNA adukata. S druge strane, fizički agensi su ultraljubičasto i ionizirajuće zračenje. Oni oštećuju DNA stvarajući jednolančane i dvolančane lomove, kromosomske aberacije, dimere pirimidina te kisikove reaktivne vrste. Unatoč tome, znanstveno je dokazana korist nekih kemijskih i fizičkih agensa u liječenju tumora. Otkrivanje mutacija vrši se različitim metodama od kojih su najpoznatije: Ames-ov i mikronukleus test, te komet test. Nadalje, Ames-ov test primjenjuje se za detekciju mutacija nastalih supstitucijom parova baza i pomakom okvira čitanja, mikronukleus testom se mogu detektirati klastogene ili aneugene mutacije, dok se komet test koristi za detekciju jednolančanih ili dvolančanih lomova na DNA, purinskih lezija i DNA adukata.
Abstract (english)
Mutations are permanent alternations of genetic material that occur in somatic or reproductive cells. They can be spontaneous, as a result of a replication error, or induced by chemical or physical agents. Chemical agents include chemical compounds that are metabolized into an electrophilic form, alkylating agents, base analogues, crosslinking agents and metal ions. They tend to cause DNA damage using various mechanisms such as base substitution, insertion or deletion of bases, and the formation of DNA adducts. On the other hand, physical agents are ultraviolent and ionizing radiation. They cause DNA damage by creating single-stranded and double-stranded breaks, chromosomal aberrations, pyrimidine dimers and reactive oxygen species. But despite the downsides, scientists were able to find their use in the form of tumor treatment. Mutation detection is done by using various methods, the most known of which are: Ames test, micronucleus and Comet assay. Furthermore, the Ames test is used to detect frame shift mutations and base pair substitution, the micronucleus assay can detect clastogen or aneugenic mutations, while the comet assay is used to detect single-stranded or double-stranded breaks, purine lesions and DNA adducts.
