Abstract
Sekvenciranje DNK, odnosno određivanje poretka nukleotida u genomu, ima važne primjene, kao što su proučavanje utjecaja lijekova i podložnosti bolestima. Tehnologijom koja je danas dostupna nije moguće sekvencirati cijeli genom, nego dobivamo podnizove od kojih ga na različite načine rekonstruiramo. Često korišten pristup su de Bruijnovi grafovi, vrsta usmjerenih grafova koje je uveo matematičar Nicolaas de Bruijn. Koristeći dobivene podnizove genoma jednakih duljina konstruiramo de Bruijnov graf, a da bi odredili njihov poredak tražimo Eulerovu stazu. U ovom radu proučena su neka svojstva tih grafova, istaknute su njihove prednosti i mane pri rješavanju problema rekonstrukcije DNK i dan je jednostavan primjer njihove primjene u programskom jeziku Python.
Abstract (english)
DNA sequencing, or the determination of the nucleotide order in the genome, has important applications, such as the study of drug effects and susceptibility to disease. With the technology available today, it is not possible to sequence the entire genome, but we obtain subsequences from which we reconstruct it in various ways. A commonly used approach are de Bruijn graphs, a type of directed graphs introduced by the mathematician Nicolaas de Bruijn. Using the obtained genome subsequences of equal lengths, we construct a de Bruiijn graph, and search for an Eulerian trail to determine their order. In this paper, some properties of these graphs are presented, their advantages and disadvantages in solving the problem of DNA reconstruction are highlighted, and a simple example of their application in the Python programming language is given.
